You can use TweetDeck - https://tweetdeck.twitter.com/ to filter tweets by keyword and handle using Twitter's search operator and then you can monitor for tweets with desktop notifications.
This solution works if you constantly monitor on desktop. But if you require email or other alerts then some other solution is needed. Besides there's rumor that Twitter might make TweetDeck a paid service.
